Melanotan-2 is a synthetic linear peptide built from seven amino acids arranged in a short chain. Its sequence is commonly written as Ac-Nle-Asp-His-D-Phe-Arg-Trp-Lys-NH2, which includes a modified N-terminus and an amidated C-terminus. The molecule belongs to the melanocortin family and acts as a receptor agonist. Structural features such as the D-phenylalanine residue and the Nle substitution are associated with increased stability against enzymatic degradation relative to the natural parent peptide.
The compound emerged from research programs in the 1980s that examined analogues of alpha-melanocyte-stimulating hormone for pigmentation and photoprotection. Investigators modified the native sequence to extend activity duration and potency. A related analogue, afamelanotide, was developed within the same broad line of inquiry and eventually gained approval in certain jurisdictions for a rare light-sensitivity condition. Melanotan-2 itself did not progress through the same regulatory route and has no approved therapeutic indication.
Analytical confirmation of identity relies on mass spectrometry, most often coupled to liquid chromatography. Reversed-phase high-performance liquid chromatography separates the peptide from related impurities and provides a purity estimate based on peak area. Electrospray ionization mass spectrometry then confirms the expected molecular mass, while tandem mass spectrometry can map the fragment sequence. For research-grade material, these two techniques together form the standard minimum. Purity figures reported by vendors are frequently not traceable to an independent laboratory.
Independent verification is central to quality control because the compound is not produced under pharmaceutical manufacturing standards. Third-party laboratories can measure purity, identity, residual solvents, and microbial contamination, though the scope of testing varies between services. Reported analyses of vendor samples have shown batch-to-batch variation in peptide content and the presence of truncated or oxidized species. How much of this variation reflects synthesis conditions versus storage and shipping is not well characterized. No harmonized reference standard exists for the material as sold.
Handling guidance for melanotan II follows general practice for small synthetic peptides rather than a product-specific monograph. Lyophilized powder is typically kept at minus twenty degrees Celsius or colder, protected from light and moisture, because warmth and humidity accelerate degradation. Once reconstituted, solutions are usually refrigerated and used within a short window, as hydrolysis and microbial growth both become concerns. Repeated freeze-thaw cycles are generally avoided. These conventions come from laboratory peptide chemistry and not from formal stability studies on this specific compound.

Melanotan-2 is frequently confused with afamelanotide, a linear analogue authorised in the European Union for erythropoietic protoporphyria. The two compounds differ in chain length, ring structure and receptor selectivity, so findings for one cannot be transferred directly to the other. Published controlled human data on melanotan-2 remain sparse, and much of what circulates online derives from small studies or unpublished reports. Questions about effect size, dose-response behaviour and long-term safety therefore remain unresolved.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone. Its sequence incorporates a lactam bridge that constrains the peptide into a ring, which increases resistance to enzymatic breakdown relative to the natural hormone. Researchers at the University of Arizona synthesised the compound in the late 1980s and early 1990s while studying pigmentation pathways. It has never received marketing approval from any national medicines regulator. In the scientific literature it is usually described as a laboratory research reagent rather than a therapeutic product.
The peptide acts as a non-selective agonist at melanocortin receptors, showing affinity for MC1R, MC3R, MC4R and MC5R. Activation of MC1R on melanocytes drives the conversion of tyrosine into melanin and shifts production toward the darker eumelanin form. MC4R signalling in the central nervous system is linked to appetite and energy balance, which helps explain why reduced food intake appeared in early human studies. Effects on MC4R and on vascular tone also account for the erectile responses recorded as unexpected findings in those same trials.
Identity and purity are assessed mainly by reversed-phase high-performance liquid chromatography with ultraviolet detection, often paired with mass spectrometry. Retention time supports identity, while the mass spectrum confirms the molecular weight of the intact peptide. Purity is frequently reported as a percentage of total peak area, a figure that depends on the wavelength, column and gradient used. Impurity profiling may also look for truncated sequences, oxidised forms and residual counterions. Amino acid analysis and peptide mapping provide orthogonal confirmation when required.
Regulatory status varies by jurisdiction, and the substance is frequently described as unapproved for therapeutic use. Some authorities classify it alongside prescription-only medicines or controlled categories, while others address it through general consumer protection rules. Analytical surveys have reported mismatches between label claims and measured content in products sold online, although the scope of such testing is limited. Whether these discrepancies are widespread remains an open question. Discussion in the literature therefore tends to combine chemistry, supply-chain observation and policy analysis.
Early published reports described melanotan-2 as a tanning agent without sun protection, which means darkening is not the same as protection against ultraviolet radiation. Later studies explored the peptide in erectile dysfunction, hemorrhagic shock, and some skin conditions. No regulator in the United States or Europe has approved it for clinical use. Many products labelled melanotan-2 are sold without approval and their identity and purity are unverified. Its long-term safety in humans remains an open question.
Melanotan-2, also written Melanotan II, is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone. Its sequence is Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, and the lactam bridge between the aspartate and lysine side chains constrains the peptide into a ring. This structural change increases receptor affinity and metabolic stability relative to the native hormone. The compound was created in the 1980s as a research tool for studying pigmentation biology.
Routine characterisation relies on reversed-phase high-performance liquid chromatography with ultraviolet detection near 214 nanometres, using a C18 column and a water-acetonitrile gradient containing trifluoroacetic acid. Electrospray ionisation mass spectrometry confirms the expected molecular mass and can reveal truncated or oxidised by-products that co-elute poorly. Sequence and stereochemistry require additional work, such as peptide mapping or amino acid analysis, because a chromatographic purity figure alone does not distinguish a diastereomer from the target peptide. Independent testing of research-grade material frequently shows measured content below the stated label, so a certificate of analysis is best read together with the method that produced it.
Melanotan-2 is handled in the laboratory as a lyophilised powder that dissolves readily in water, dimethyl sulfoxide and dimethylformamide, with limited solubility in ethanol. Stock solutions prepared in an organic solvent often precipitate when diluted into aqueous buffer, so gradual dilution with mixing is standard practice. The peptide carries a tryptophan residue and a histidine residue, both sensitive to oxidation and to alkaline conditions. Working solutions are therefore kept near neutral to slightly acidic pH, protected from light, and consumed within the same working session whenever that is practical.
Methylmalonyl-CoA is the thioester consisting of coenzyme A linked to methylmalonic acid. It is an important intermediate in the biosynthesis of succinyl-CoA, which plays an essential role in the citric acid cycle.

=== Recreational use === Zopiclone has the potential for non-medical use, dosage escalation, and drug dependence. It is taken orally and sometimes intravenously when used non-medically, and often combined with alcohol to achieve euphoria. Patients abusing the drug are also at risk of dependence. Withdrawal symptoms can be seen after long-term use of normal doses even after a gradual reduction regimen. The Compendium of Pharmaceuticals and Specialties recommends zopiclone prescriptions not exceed 7 to 10 days, owing to concerns of addiction, tolerance, and physical dependence. Two types of drug misuse can occur: either recreational misuse, wherein the drug is taken to achieve a high, or when the drug is continued long-term against medical advice. Zopiclone is as addictive as or may be more addictive than benzodiazepines. Those with a history of substance misuse or mental health disorders may be at an increased risk of high-dose zopiclone misuse. High-dose misuse of zopiclone and increasing popularity amongst people who use substances who have been prescribed with zopiclone was shown in the 1990s. The symptoms of zopiclone addiction can include depression, dysphoria, hopelessness, slow thoughts, social isolation, worrying, sexual anhedonia, and nervousness. Zopiclone and other sedative hypnotic drugs are detected frequently in cases of people suspected of driving under the influence of drugs. Other sedating drugs, including benzodiazepines and zolpidem, are also found in high numbers of suspected drugged drivers.
=== Batch === In a batch process, all the ingredients are combined and the reactions proceed without any further input. Batch fermentation has been used for millennia to make bread and alcoholic beverages, and it is still a common method, especially when the process is not well understood. However, it can be expensive because the fermentor must be sterilized using high pressure steam between batches. Strictly speaking, there is often addition of small quantities of chemicals to control the pH or suppress foaming. Batch fermentation goes through a series of phases. There is a lag phase in which cells adjust to their environment; then a phase in which exponential growth occurs. Once many of the nutrients have been consumed, the growth slows and becomes non-exponential, but production of secondary metabolites (including commercially important antibiotics and enzymes) accelerates. This continues through a stationary phase after most of the nutrients have been consumed, and then the cells die.

=== Research === TRH has been evaluated for the treatment of various neurological disorders. It has been attempted for treatment of various epileptic disorders. TRH has been shown to improve outcomes of CNS injuries in experimental models. Efficacy for the treatment ALS and spinal muscle atrophy has not been demonstrated. Many individuals with depression exhibit a blunted endocrine response to TRH due to unknown reasons, and the response is correlated with clinical outcomes. Involvement of TRH in the pathogenesis of depression has nevertheless not been well established. TRH has undergone research for its ostensible antidepressant properties, however, results regarding efficacy have been inconsistent. One study on a small sample of people with treatment-resistant depression found short-lived anti-depressant and anti-suicidal effects when TRH was administered intrathecally. An orally bioavailable prodrug is being researched. In 2012, the U.S. Army awarded a research grant to develop a TRH nasal spray for suicide prevention amongst veterans. TRH acts as a wakefulness-promoting agent, causing awakening from sleep or sedation.
Title II of Act, the Controlled Substances Act (CSA), helped align US law with the UN Single Convention, with "many of the provisions of the CSA ... enacted by Congress for the specific purpose of ensuring U.S. compliance with the treaty." The CSA's five drug Schedules, an implementation of the Single Convention's four schedule system, categorized drugs based on medical value and potential for abuse. Under the new drug schedules, cannabis was provisionally placed by the administration in the most restrictive Schedule I, "until the completion of certain studies now underway to resolve the issue." As mandated by the CSA, Nixon appointed the National Commission on Marijuana and Drug Abuse, known as the Shafer Commission, to investigate.

Spondyloepiphyseal dysplasia congenita is one of a spectrum of skeletal disorders caused by mutations in the COL2A1 gene. The protein made by this gene forms type II collagen, a molecule found mostly in cartilage and in the clear gel that fills the eyeball (the vitreous). Type II collagen is essential for the normal development of bones and other connective tissues. Mutations in the COL2A1 gene interfere with the assembly of type II collagen molecules, which prevents bones from developing properly and causes the signs and symptoms of this condition. Spondyloepiphyseal dysplasia congenita is inherited in an autosomal dominant pattern, which means one copy of the altered gene is sufficient to cause the disorder.
Gabapentin is a 3,3-disubstituted derivative of GABA. Therefore, it is a GABA analog, as well as a γ-amino acid. It is similar to several other compounds that collectively are called gabapentinoids. Specifically, it is a derivative of GABA with a pentyl disubstitution at the 3 position, hence, the name - gabapentin, in such a way as to form a six-membered ring. After the formation of the ring, the amine and carboxylic groups are not in the same relative positions as they are in the GABA; they are more conformationally constrained. Although it has been known for some time that gabapentin must bind to the α2δ-1 protein to act pharmacologically (see Pharmacodynamics), the three-dimensional structure of the α2δ-1 protein with gabapentin bound (or alternatively, the native amino acid, L-Isoleucine bound) has only recently been obtained by cryo-electron microscopy. A figure of this drug-bound structure is shown in the Chemistry section of the entry on gabapentinoid drugs. This study confirms other findings to show that both compounds alternatively can bind at a single extracellular site (somewhat distant from the calcium conducting pore of the voltage-gated calcium channel α1 subunit) on the calcium channel and chemotaxis domain of α2δ-1.

=== Origins (1992–2004) === Julien Rour Chanut grew up in the Essonne department, south of Paris, in France. Mehdi Birouk Thépegnier, born in Morocco (Maghreb), arrived in France at the age of 12. They met in 1992 at a college in Essonne and became friends. Chanut and Thépegnier were passionate about 1990s hardcore punk, heavy metal and grunge. The hardcore scene consisting of bands such as Confusion, Integrity, Only Living Witness, Section 8, and Sheer Terror, and the do-it-yourself philosophy associated with this culture, motivated them to perform music. Chanut found his path between the ages of 13 and 14 by beginning to play hardcore punk. The two friends first stepped onto the stage at around 15 or 16. They grew up in the hardcore punk scene of the 1990s. Thépegnier was a member of Mouvement Colère (lit. 'Anger Movement') and later a bassist in Drowning, which toured as the opening act for All Out War and Merauder. Over time, Chanut and Thépegnier performed in various local bands, including Arkangel (band), Es La Guerilla and Rising Dust. Clément Hanvic was part of their circle; he was the singer of Knockoutz and the bassist of L'Esprit du Clan (lit. 'The Spirit of the Clan'), and performed with Es La Guerilla. The bassist for Knockoutz and Es La Guerilla was Kéo Nackphouminh. Sid-Ahmed Azzouni was a guitarist in Es La Guerilla. After playing electric bass for a while, Thépegnier switched to drums.
In response to the potential ban, many users signed up for the Chinese app Xiaohongshu (Chinese: 小红书), known internationally as REDnote. On 18 January 2025, hours before the bill went into effect, TikTok became unavailable across the country. The next day, TikTok restored access to their service after re-elected US President Donald Trump assured TikTok he would not enforce the law. President Trump signed an executive order on 20 January 2025, delaying the enforcement of the TikTok ban by 75 days. In April 2025, President Trump signed another executive order further delaying the enforcement of the TikTok ban by 75 days, and did so again in June 2025 with a 90-day extension. The Department of Justice's Office of Legal Counsel determined that TikTok USDS, the version of TikTok operated by the TikTok U.S. Data Security Joint Venture, was not subject to the No TikTok on Government Devices Act, thereby permitting employees to use TikTok on government devices, subject to agency discretion and workplace policies.
=== Crosstalk with Other Metals === hCTR1 is highly selective for copper due to the rich methionine transmembrane selectivity filter, copper homeostasis does not operate in isolation. One well documented but still controversial direct interaction is with zinc. The interaction between zinc and copper is highly complex. Zn(II) can antagonize copper uptake in vivo, high zinc possibly induces a functional copper deficiency. Oral zinc therapy is used to treat Wilson’s disease patients by blocking intestinal copper absorption. There are four principal levels of crosstalk: intestinal absorption, the transcriptional regulation of SLC31A1, direct interactions with hCTR1 and zinc-dependent modulation mediated by Zinc Transporter 1. Another important crosstalk is silver. Ag(I) is chemically similar to Cu(I), both are monovalent “soft” cations, and silver can enter cells via hCTR1. Experimentally, silver ions have been shown to inhibit copper uptake by blocking hCTR1. Met43 and Met45 within the N-terminal domain are key residues responsible for Ag(I) coordination. This cross-reactivity has practical implications, for instance silver-based antimicrobials could disrupt human copper homeostasis by competing for hCTR1. Furthermore dietary silver or high copper concentrations could interfere with zinc and iron metabolism due to shared regulatory networks. Copper homeostasis also intersects with platinum, which is very well documented in the context of chemotherapy. The commonly used platinum drug, cisplatin enters cells partly via hCTR1.
== History == The effectiveness and safety of apitegromab were evaluated in a 52-week randomized, double-blind, placebo-controlled trial (NCT05156320). The trial enrolled 188 participants with spinal muscular atrophy between 2 to 21 years of age who were not able to move or walk independently. All participants were already receiving an approved survival motor neuron 2-targeted treatment. Participants were randomly assigned to receive apitegromab 10 mg/kg, apitegromab 20 mg/kg via intravenous infusion, or placebo once every four weeks for approximately one year. The primary analysis was conducted in 156 participants aged 2 to 12 years of age.

No. Melanotan-2 is manufactured synthetically. The naturally occurring peptide in the same family is alpha-melanocyte-stimulating hormone, which the body produces as part of normal endocrine and neural signalling.
Both are synthetic analogues derived from the same parent hormone. Reference descriptions usually characterise afamelanotide as more selective for the MC1R subtype, whereas melanotan-2 is reported to interact with a wider range of melanocortin receptors.
Early studies explored melanocortin signalling and pigmentation, which later led to informal use of the label tanning peptide. That label is not a regulatory category and does not imply approved status for any purpose.
The lyophilized powder is generally held at minus twenty degrees Celsius or below, away from light and moisture. Reconstituted solutions are typically refrigerated and used quickly. These practices derive from general peptide handling rather than a formal stability study.
